METHOD

Solvent: 10 mM Potassium Phosphate/H2O, pH 7.3

Infrared Measurements:Nicolet 730, Magna 550, or Bomem-MB Fourier transform Infrared Spectrometer; Single beam mode with a 4 cm-1 resolution at room temperature.

Subtraction of Water Spectra:

Criterion I: The bands originating from water vapor must be subtracted accurately from the protein spectrum in the 1800 to 1500 cm-1 region regardless of the baseline (gaseous water subtraction).

Criterion II: A straight baseline must be obtained from 2000 to 1720 cm-1 (aqueous water subtraction).
